You need also to do so for city-states else they sometimes want to start next door to your starting position or next-door to one of the other major civs depending on whether the game decides a particular area is a 'sweet-spot' for civs and city-states to spawn at. You will also want to assign the othere civs starting points by specifying random player starts for all the other civs, or designating which civ will start in each of the player start positions you designate. The drop-down in Edit Plots is the one that says 'start position'. After you define the civs you want used in the scenario, you go back and use the Edit PLot tab to select Player1's starting position at whichever tile (plot) you wish to use.
You have to make the map a scenario by adding the civs you want used in the scenario (scenario editor menu in the middle upper area of the Worldbuilder taskbar).
